RESUMO

Species with extensive geographical ranges pose special challenges to assessing drivers of wildlife disease, necessitating collaborative and large-scale analyses. The imperilled foothill yellow-legged frog (Rana boylii) inhabits a wide geographical range and variable conditions in rivers of California and Oregon (USA), and is considered threatened by the pathogen Batrachochytrium dendrobatidis (Bd). To assess drivers of Bd infections over time and space, we compiled over 2000 datapoints from R. boylii museum specimens (collected 1897-2005) and field samples (2005-2021) spanning 9° of latitude. We observed a south-to-north spread of Bd detections beginning in the 1940s and increase in prevalence from the 1940s to 1970s, coinciding with extirpation from southern latitudes. We detected eight high-prevalence geographical clusters through time that span the species' geographical range. Field-sampled male R. boylii exhibited the highest prevalence, and juveniles sampled in autumn exhibited the highest loads. Bd infection risk was highest in lower elevation rain-dominated watersheds, and with cool temperatures and low stream-flow conditions at the end of the dry season. Through a holistic assessment of relationships between infection risk, geographical context and time, we identify the locations and time periods where Bd mitigation and monitoring will be critical for conservation of this imperilled species.

Kinetochores are smaller and more accessible experimentally in budding yeast than in any other eukaryote. Believing that simple and complex kinetochores have important structural and functional properties in common, we characterized the structure of CBF3, the essential centromere-binding complex that initiates kinetochore formation in Saccharomyces cerevisiae. We find that the four subunits of CBF3 are multimeric in solution: p23(Skp1) and p58(Ctf13) form a heterodimer, and p64(Cep3) and p110(Ndc10) form homodimers. Subcomplexes involving p58 and each of the other CBF3 subunits can assemble in the absence of centromeric DNA. In these subcomplexes, p58 appears to function as a structural core mediating stable interactions among other CBF3 proteins. p58 has a short half-life in yeast, being subject to ubiquitin-dependent proteolysis, but we find that it is much more stable following association with p64. We propose that p23(Skp1)-p58-p64 complexes constitute the primary pool of active p58 in yeast cells. These complexes can either dissociate, reexposing p58 to the degradation pathway, or can bind to p110 and centromeric DNA, forming a functional CBF3 complex in which p58 is fully protected from degradation. This pathway may constitute an editing mechanism preventing the formation of ectopic kinetochores and ensuring the fidelity of chromosome segregation.

RESUMO

Debate continues over the use of compartment pressure monitoring devices in the diagnosis of compartment syndrome. We present the results of a survey that was designed to explore this issue. The majority of UK trauma and orthopaedic surgeons surveyed advocate making the diagnosis of compartment syndrome by a combination of clinical acumen and compartment pressure measurements. Less than half of the trauma centres surveyed had pressure monitoring equipment available. There is marked variation in opinion regarding the threshold level for surgery when monitoring is applied.

RESUMO

Intravenous injection of the oral formulation of Temazepam is a common form of drug abuse. At the request of the Home Office, manufacturers have developed solid dose forms to replace the liquid filled capsules, but a number of cases of accidental intra-arterial injection of the new formulation have been reported, with varying management strategies advocated. A case is presented where prompt aggressive management using surgical and pharmacological measures resulted in complete limb salvage. Previous reported cases and management options are discussed.

The four nucleolar proteins NOP1, SSB1, GAR1, and NSR1 of Saccharomyces cerevisiae share a repetitive domain composed of repeat units rich in glycine and arginine (GAR domain). We have cloned and sequenced a fifth member of this family, NOP3, and shown it to be essential for cell viability. The NOP3 open reading frame encodes a 415 amino acid protein with a predicted molecular mass of 45 kD, containing a GAR domain and an RNA recognition motif. NOP3-specific antibodies recognize a 60-kD protein by SDS-PAGE and decorate the nucleolus and the surrounding nucleoplasm. A conditional lethal mutation, GAL::nop3, was constructed; growth of the mutant strain in glucose medium represses NOP3 expression. In cells depleted of NOP3, production of cytoplasmic ribosomes is impaired. Northern analysis and pulse-chase labeling indicate that pre-rRNA processing is inhibited at the late steps, in which 27SB pre-rRNA is cleaved to 25S rRNA and 20S pre-rRNA to 18S rRNA.

Turkey poults were suffering from diarrhea on a farm in which several previous grow-outs of turkeys had experienced a clinically identical problem. Upon necropsy, significant gross lesions were restricted to the gastrointestinal tract. Segments of small intestine were pale and distended with cloudy mucoid material and a few gas bubbles. The ceca contained fluid and gas. Fresh organ portions were collected and fixed in 10% neutral buffered formalin and submitted for histological processing and examination (light microscopy and scanning and transmission electron microscopy). Small (2-4 micron) basophilic bodies identified as Cryptosporidium sp. were present in enterocytes of the middle and lower small intestine. The villi were moderately atrophic, the crypts were hypertrophic, and the lamina propria was infiltrated by large numbers of lymphocytes, heterophils, and fewer macrophages and plasma cells. Numerous intraepithelial leukocytes and exocytosing inflammatory cells also were present.

Groups of broiler chickens were fed rations containing .625, 1.25, or 2.5 micrograms total aflatoxin/g of feed from one day to 42 days of age. Treatment groups were segregated according to sex and control groups were maintained. The chickens were bled at 14-day intervals, and serum was collected for assay of total complement (C) activity using a radial hemolytic procedure. No significant differences were found in C titers between males and females of the same age group. For the sexes combined, mean C titers of chickens fed aflatoxin were compared to mean titers of controls. For each age group, significant differences in C titers existed only between the control group and those chickens fed 2.5 micrograms/g aflatoxin. Complement is a major component of the humoral immune response and this study adds to the evidence that aflatoxin can be immunosuppressive to chickens.

Four commercial disinfectants used in poultry house sanitation procedures were evaluated for efficacy using a specialized petri dish (Rodac plate) for contact sampling. The samples were taken from a variety of surface materials commonly found in poultry rearing facilities. Two media were used, one for bacteria and one for mold. Predisinfection samples were taken following removal of chickens and litter. Postdisinfection samples were taken 4 hr after room treatment with a high pressure spray apparatus. The most effective disinfectant for reducing bacteria colony counts was a product that contained glutaraldehyde. The other products, ranking in order of efficacy for bacteria, contained cresylic acid, iodophors, and a combination of quaternary ammonium compound and formaldehyde. The most effective disinfectant for reducing mold colony counts was the product containing cresylic acid. The other products, ranking in order of efficacy for mold, contained iodophors, the combination of quaternary ammonium compound and formaldehyde, and glutaraldehyde. The most effective overall disinfectant was the cresylic acid product.

Broiler chickens and turkeys were tested for hemolytic complement (C) activity at 7 or 14 day intervals beginning at one day of age and ending at 42 days of age. The mean titers for chickens on days 1, 14, 28, and 42 were 1:14, 1:14, 1:22, and 1:33, respectively. The mean C titers for turkeys on days 1, 7, 14, 21, 28, and 42 were 1:4, 1:6, 1:9, 1:10, 1:19, and 1:25, respectively. Birds were segregated as to sex so hemolytic activities could be compared. There were no significant differences between sexes in C titers in broiler chickens at any of the ages tested. This was also true of turkeys, except at 42 days of age when the C titer of the hens was significantly higher than the toms. Complement titers increased with age in both broiler chickens and turkeys.
